Evaluation of two Immune-Enzimatic Assays (ELISAs) for detecting Anti-Map anti-bodies in domestic ruminants

The objective of the work was to evaluate two ELISA’s for detection anti-Map antibodies. 460 serum and fecal samples bovines (n=267), ovines (n=140), goats (n=53) were evaluated. Serums used detecting antibodies using different ELISAs, one in-house ELISA made with low-molecular weight proteins (<100 kD) from antigen ultra-filtrate Map strain 3065, a commercial paratuberculosis diagnostics. DNA extracted faeces carry out PCR-N IS900. Sensitivity (Se), specificity (E) Kappa (K) calculated estimate concordance between tests. low-weight map showed Se 87%, E 96% K 0.85, while Se=55%, E=97% K=0.576, when compared IS900 test. When comparing both 50%, 97%, K= 0.393. <100 kD had better than ELISA; them low. Low-weight obtained 3065 are good candidate as in diagnosis Johne’s disease Key words: Paratuberculosis, ELISA, diagnostic.
